Introduction: CLL is the clonal expansion of mature CD5/CD23/sIg lymphocytes. Most B-CLL cells express on their surface surface IgM or IgD, both of which function as B-cell antigen receptor (BCR). The majority of CLLs originate from antigenexperienced, memory B cells with mutated variable region (V) genes (M-CLL) and have a more indolent course. CLL cells with unmutated VH genes (U-CLL), more aggressive, might derive from B-cells in which antigenic stimulation did not result in somatic mutations. The prognostic difference between M-CLL and U-CLL might originate from differences in responsiveness of the mutated or unmutated BCR to stimulation. We interrogated signaling pathways in primary CLL cells using a novel multi-parameter flow cytometric technique.
